The first step we will be taking is choosing our team. You are going to wanna pick Ayane and Xu Shu for this and hopefully after all your previous playing you have 3-4 promotions in each of them. As far as the rest of your team goes thats 100% up to you. Personally I chose Sterkenburg, Orochi and Orochi X to build myself a team of powerhouses.
As far as gauntlet mode itself goes the mode is basically the entire game with harder enemies and levels and lots lots lots lots more items. The longer you stay in a level the more difficult it becomes. The miasma level increases based on time spent in the level and enemies + officers killed. With Ayane and Xu Shu you can easily handle 100- 120 level miasma. By this point you are going to want to have Windslay, Slay, Agression, Courage, Osmosis, Destruction, Agility and Alacrity. Using these abilities will not only give you a power boost every 200 combo but every 500 will also completely heal your team.
If you run out of musou with Ayane simply switch to Xu Shu and build a quick 500 combo then switch back. With many promotions under your belt you can easily move faster than red hare and all other horses in the game. Once you hit 5-6 promotions assuming you saved all your growth points you should now be able to max out all the stats of Ayane and Xu Shu. The basic tactic you will use will be the hit and run. Run from mob to mob and simply use if playing as Ayane or if on Xu Shu. Using these methods you should have absolutely no problems clearing levels at all.
A little about levels. There are 4 different types of level colors.
Red, Blue, Yellow and Purple. By simply clearing a level you gain one crystal. At the beginning the only levels you can play are the red crystal levels. Upon completion of gauntlet mode (all 4 levels) you unlock the next crystal color and must do it again. In the beginning don't worry about doing every single stage since you need to clear the mode a total of 4 times. Simply do a blitz to the exist until you get to your 4th run then clear every level as you will to.
Doing so will unlock. IMPORTANT There is an extremely OP formation that makes clearing stages literally instant. Its called the solstice formation. This formation is unlocked at 95 ally unlocks. Until you get this formation the demon eye formation unlocked at 60 shows the exit which is still very helpful. For your first few levels you should try to unlock as many ally officers as possible.

As far as the other named officers go just be on the look out for stages that contain their name in them. Kill them then use solstice or demon eye to exit the level. For farming 500 of each type of unit there really isn't a great tactic other than trying to get only 1-2 unit types in a single stage.
OR you can shoot for a single stage that has 3-4 that you do not have done and farm that numerous times. I would suggest always keeping a total unit kill request at all times and a general officer killing one along with a famous officer request. As far as farming the otherworldly invaders. Starting up purple stages with red names and waiting for 5-10 chests to turn red is a fantastic way to crush this grindy request. If you happen to get poor stage units you can re-roll a stage for 5 of whatever crystal.
I would suggest doing this often with the reds as you will have quite a lot of those considering you need 4 runs minimum. Save your other three colors for the higher tier unit kills. Make sure to save before using them all to avoid using all your crystals.
